You can try to rebuild your netinfo database. This will wipe out the existing db and install a fresh copy. It should get you back to a functional state with Netinfo manager.

Boot into single user mode (command - s)
Do an fsck to clean up any disk problems first:

fsck -y

Mount drive:

mount -uw /

Then get to the netinfo database and rebuild:

cd /var/db/netinfo
mv local.nidb local.nidbold
nicl -raw local.nidb -create

Here is a more complete Apple procedure if you want to restore one of the auto-backups of Netinfo:

I'm thinking a reinstall of OSX. You can try the archive install where it preserves your user space first and if you still have problems a reformat install is probably best.

What have you tried so far? I am assuming you fired up NetInfo Manager ->Security->Authenticated then tried to enable the root user?

Have you tried Disk Utility -> Repair Disk/Repair permissions?

I would save Archive and Install as the measure of last resort though. It takes me some time to manually move stuff I need from the archived /Library to the new /Library. BTW, why do you need to enable root? Is there something you can't sudo your way though?
