What size screen? 1024 x 768? 800 x 600? Who's the audience and what do they expect to see. What do you want to display? These are all questions that will affect the path you go down - if you have an information-heavy site then it may be a silly idea to constrain it all to the size of an 800 x 600 screen, but if you are promoting your video clips then that may be all you need.
