There are a few programs out there that allow you to define
short cut key (or hot key) for lauching other program.
Is there API provided by OSX to do these? I'm always curious
how those program can capture the key event when certains key
is pressed. I've been looking around and couldn't find
any info on this topic at all.

I take it you mean like Snapz Pro X does (invokes when cmd-shift-3 is pressed). There is an API for doing so in Carbon. It needs your application to be running (faceless and in the background).
