Is there a way to update the iTunes play count and last played date from an iPod? In other words if I play a song on my iPod, the play count and last play date are changed to today on my iPod, I'd like this to be updated in iTunes library for the same song. Is this possible?

I've just checked, and mine actually does do this.

Im pretty sure it used to when the iPod first came out, then I think it stopped doing so, but it seems to be working now, might be the iT4.2 update or something.

I think this is true of the 3G iPods, but not earlier models (such as mine).

Mine is an earlier model.

I remember it working when i first got the iPod, then am sure it hasn't worked for months, but seems to be working now.

I have a new 10 gig, so I'm assuming it as a 3G (dock connector). I have iTunes set to manage the iPod manually. Does it need to be set to automatically sync or something?

In my quick test, it does update if I have the iPod auto-sync a playlist. However, I want to be able to manually put various (not always the same) playlists on my iPod, and then have the sync work with a manual update. Any suggestions?

I have mine to auto sync 2 smart playlists with what i most/recently listen to. and then have a playlist called "To iPod" that i drag stuff to manually.

You could do that, just have it set to auto sync a single playlist, and drag what u want to there.
