to audio conference through iChat, do you have to have a phone line? (i.e. dsl, or modem)
or can it be done through a cable connection?

I see no reason why it shouldn't work with cable.

That's great to know ... I'm preparing to set my buddy up with his first Mac. He'll be on dialup and I was hoping I'd be able to audio chat with him (I imagine I'll be doing quite a bit of tech support since this is both his first Mac AND first computer since his eMachines died nearly 2 years ago). The guy doesn't type ... he hunts and pecks at about 2 words per minute. The thought of tech-supporting him by typing was a terrifying thought

I have cable at home and dial-up at the office. Audio chat works fine in both places. Just keep in mind that for now, audio chatting is Mac-to-Mac only.
