I've used the iTunes Music store since it opened and never had any problems until about two weeks ago, now I get constant -50 errors and I can't download my music, plus I don't get full previews I only get maybe 10 or 15 seconds of preview then it cuts off.
Very frustrating.
I got the error with 3 different Macs, thru two different ISPs. I got the error with both iTunes 4.7 and 4.7.1. Got the error on a Windows box as well.
Others seam to be having the same problem with no luck on fixing it, it seems like an Apple / Akamai problem. I've even had problems downloading other content from Apple's site as well like movie trailers or watching the MWSF Keynote stream and in the past I've never had any problems with Apple's site.
If you have any of these problems please report them to Apple, it may help them find a fix.

Apple is working with Akamai on the problem. Akamai appears to be having some serious problems with their distribution network - they also have had problems with the MacWorld keynote stream - and it's taking time to work out the cause and arrive at a fix.
Hang in there. I know it's frustrating, but the issue is being worked on. The problem at this point, though, appears to be largely out of Apple's hands but rather up to Akamai to fix.
