I've tried to add artwork to MP3s not purchased from iTunes, but when I click "Add Album Artwork" nothing happens.

I also have Download Artwork selected in the Preferences.

Nothing happens - no beach ball, no iTunes store download bar.

Nothing is supposed to happen. If iTunes can find the artwork, it uses it. If it can't, nothing happens.

Try Art Collector

Make sure that:
1) All your ID3 tags are correct (Album, Year, Genre etc.)
2) That the album is actually available in the iTMS
3) That you have an iTMS account
