Create a new message in Mail...from the "Message" menu, choose "Mark" and then choose "As High Priority"...

If you do this frequently, you can add it to your compose window as a menu:

You can also add the "Flags" column under the View > Columns menu as well so you can see when you receive flagged/high priority messages.
