This has been happening a lot recently. My Sound Sticks will be working fine, and then all of a sudden I get no sound. What's more interesting is that if they're not working and I try to play something in iTunes, it just sits there on Pause and won't play the song. The built-in audio works fine, the problem is just with anything else I try to use.

Is there something I can trash somewhere and make everything all better?

Sleep the computer and wake it back up. My solution is unplugging my iMic (which is connected to my amplifier) en reconnecting it. Maybe deconnect and reconnect the Sound Sticks (are they USB ? I think so.. not sure actually)

Just for future reference (if needed), the sound pref file is located in:
/Library/Preferences/com.apple.soundpref.plist
Note: That's your main system Library folder, not your Home Library.
