Since loading Panther on their machines, two of my users have had random lock-ups, incomplete boot processes...the system gets to a certain point...different points...and locks up, lines on the screen, and complete blackouts like the machine has gone to sleep and won't wake up.

One of the machines is an iMac 800, the other a Titanium Powerbook. Both ran 10.2 fine, and both loaded Panther over 10.2...did not do a clean install.

Should we do a complete reformat and start over? Is it something else?

I would try archive and install first as that will not overwrite your data. Of course back-up first. If that does not work a clean install is probably the next thing to try.
