Am running 10.4.1 and Quicktime 7.01.

Whenever I try to play the WWDC keynote (or in fact any video from the Apple Events section of the Quicktime website) I get no sound. I can play other files (trailers etc.) with no problems. Any other local movies etc work fine.

When I try to run Virtual PC I get this message:
"Virtual PC is unable to play sounds. VPC is unable to start the mac audio system. A newer version of the Mac OS might fix this problem".

I have created a new user and the problem persists in the new user.

I have done an archive and install and this doesn't resolve the problem either.

Anyone know where the problem might be and how I can fix it?

Do you have a sound card? Do you have any remotely related sound equipment connected to your machine (even the iSight counts here)? What kind of machine do you have?
