Possibly try a verbose start up. Command - v has for some reason worked its magic on me, and it might also tell you where theings are going wrong.

Have you also tried a safe boot by holding shift at start up?

All is not lost yet.

And being a seasoned professional, you do have back-ups, right?

Pressing option command shift and delete during start-up should allow you to boot from another source according to a page I have in front of me, I thought it was just C, but according to this page (from apple) that is for CD's.

Also X during startup is for a force startup.

This page I printed out about 3 years so whether it is still 100% correct.....

It is more about where it stops.

What did DU tell you, and do you have DW? Maybe no need for a trip to the store at all, though a more knowledgeable person might breeze through and have a more informed assessment.
