Best way to add a new drive?

I need to add more HD space. What is the best way to configure a new internal drive under OSX? Can I mount the new drive as a subdirectory of my home folder so that everything that goes on it is under my permissions or do I have to mount it at the computer level? Any advice would be appreciated.
