What does using DarWINE need?

What does DarWINE need to be present in the system besides X11? I managed to foul it up so thoroughly that I thought I might as well give up on trying to fix it and just reinstall everything, including the OS, from scratch. To avoid that kind of mess again, what needs to be safeguarded to keep WINE working?
Is there any software known to interfere with it?
