Backing up with Time Machine 2 drives to one?

I have 2 drives I use for apps and documents on my Mac Mini,

80Gb and 400GB

Can I use another say 500GB and back up both of these 2 drives into one external FW drive using Time Machine?

That's how Time Machine works, pretty much. By default, it will back up ALL the volumes connected to your Mac to the designated backup disk. You can configure this in the System Preferences.
