I'm looking to buy a 12" PB. If I were to make the purchase at one of Apple's retail stores, would they allow me to open the box/test the unit there and exchange it if there are major problems?

You can open the box after you purchase it, but not before.

If there are serious problems, like DOA or spots they will exchange, otherwise you are paying the 10% restocking fee (like if you had 1 dead pixel, you would lose 10% if you returned it).
