I have a HD that I rarely use, and it constantly is spinning, and when it goes to sleep, it will spin up with the other drive (which I do use)....

I know I can put the drive in the trash, and then it will be available on restart.... but is there a way to remount the drive without restarting? There are times I want the drive, but for the rest, it would be nice to get rid of it.

Under Mac OS 9:

Make an alias to the disk before unmounting it. To remount it, open the alias.

Under Mac OS 9, if that doesn't work:

Choose the Mount Volumes option in Drive Setup.

Under Mac OS X:

Use Disk Utility's Mount option with the disk(not drive) selected.

None of these will work if the drive is physically disconnected from the computer.

If you go into Volume Setting in Drive setup you can set the drive so it doesn't automount then you won't have to unmount it on bootup but you can still use the alias to mount it if you need to.
