Without a doubt, Jurassic Park. And of course, I love the theme from Superman. Hannibal has a great score as well.

I liked the soundtrack to 28 Days Later – especially the edited version of Godspeed You! Black Emperor's song. It really made the movie in my opinion.
The movie Moonlight Mile also had a pretty good soundtrack. Coen brothers movies have good soundtracks; so did The Royal Tennenbaums and The Life Aquatic.

I've read the wiki entry on it and Once Upon a Time... and I'm left feeling there's a lot of films I should be watching to further appreciate them.

There are lots of great films out there with some very unique soundtracks.
You might like the Zatoichi series as well (the one with Shintarô Katsu). It's essentially about a blind swordsman in the Edo period that avenges wrong doers.
