This is rather weird.
I could use Software Update prior to the 10.0.1 update without so much of a hickup. I was able to use it twice -- first to update Software Update and to install Mac OS X 10.0.1. But after the install/reboot, Software Update now refuses to run, saying that I should check my configuration.
I have checked my network settings and they don't seem to be wrong (since I didn't change them), and I'm very well sure I have a connection to the net since I can surf, email, etc.
Any advice is greatly appreciated.

I was the first to post this topic here. I rebooted again because I went back to OS 9 to use VPC. Software update works fine now. I forgot the responses I got so you can do a search for something like "10.0.1 killed my software update." I don't exactly remember so just look for software update or reboot.

I had the problem and deleting software updates prefs (in ~/Library references) was the solution !
