Is it possible to easily roll back to previous system upgrade. We ugraded today to 10.4.3, but something must have gotten corrupted during the process as our file sharing capabilities are nil now on one of the 10.4.3 machines. (fine on another one..)

Is it easy to roll back, or do I have to 'restore from disk' and build it back to 10.4.2?

No, but you should try installing the update using the combo updater first. Get it here. Then restart, and if sharing still doesn't work, check the logs to see if any errors were reported by the service. What kind of sharing are you running?

running very basic serving. just sharing folders from the desktop. We've changed the permissions to allow everybody read/write on the folder we're trying to share. Very strange. The other machine we upgraded to 10.4.3 works flawlessly.

Restore from disk might be overkill -- do an "archive and install" in which you preserve user accounts/settings to get back to 10.4, and then the combo updater to get to 10.4.2.

You compare it to your other machine. Do you have the exact same software on both? Is this the only thing you seem to be having trouble with?
Are there any haxies or software modifying apps on the one you are having trouble with?

If so, disable them or remove them. Reinstall the combo updater and then one by one re-enable your other software. Get updates from macupdate if necessary.

I doubt both machines are equal.

To share directly from the desktop you must be using something like Sharepoints, so make sure it's settings are correct. You may also want to try reinstalling it.

Could try repairing permissions using the Disk Utility software. That may help.

Also try making new folders and seeing if they will work for file sharing.
If I had a signature, it would look something like this
