Does anyone know how to make the scroll-wheel button open a new window (or tab) with a hyperlink?

I don't think you can do it via about:config. You'll need to set your middle mouse button to command-click with your mouse's drivers.

Firefox OS X can't do this by itself yet..
A bug has been filed on this here. Go vote for it!
