Not the most original, but Cornwall and the South Coast. The Channel Islands.

I lived in London for too long, but if you're a first time visitor, that would be on the map.

Ireland?

If you're into castles and history and Patrick McGoohan you can do North Wales. It can suck if the weather's bad though. Stay away from Rhyl, and areas of Colwyn Bay are pretty dire.

I've got Snowdonia and Portmeirion on the agenda (a day trip).
London (and all the usual contained therein), is obviously on.

Chester/Bath/Warwick/Stratford/Longleat/Henge/Jodrell/Ironbridge/BlueJohn... ...trying to rack my brain for other stuff.
Been inclined to wander... off the beaten track.

I lean towards the ancient. Some of my favorite sites include:

Uffington White Horse
Avebury Stone Circle, Silbury Hill and West Kennet Long Barrow
Drive-by of Stonehenge- look at it though the chain link to cross it off the list
If you have any thoughts of N. Ire, Boa Island and Giant's Causeway are great (though G.C. is natural)

Some of my other favorite places:
The Tan Hill Inn, highest Pub in England and bursting with character
The Minack Theatre in Cornwall (can cross Land's End off the list if you care)

This is off the top of my head, and skews towards the South and West as that's where I am. If any other inspiration strikes, I'll let you know.

We are right around the corner from The Queen to Be's hometown and if by some unexpected oddity I can give a list of Kate sites to visit if it appeals.

Also:

The Ashmolean and Pitt-Rivers museums are really good if you were thinking of visiting Oxford.

The picture above reminded me of St. Michael's Mount if you do make it to Cornwall. I would also consider, if you don't already have one and based on your planned itinerary, getting a National Trust and/or English Heritage membership. Discounts and free parking to a lot of the sites. It will also make you a card carrying member of the Big Society.
